How significant is the difference between an intel core 2 quad Q9450 2.66 GHz 12 MB cache Processor with an Nvidia 780i Core 2 Quad MoBo and an AMD Phenom 9550 2.2 GHz Quad Core 4 MB Cache Processor with an Nvidia 750a SLI MoBo? The first setup is about 300$ more expensive. Is it worth it?

Yes.  The AMD Phenom's, in my opinion at least, aren't worth it if you are looking for a high-end gaming machine. 
 
I can also tell you the 9450 overclocks VERY well; getting less then 3.0 GHz overcloked would be a dissapointment. Not sure how AMD's new chips fare though...

If you're on a tight budget, the AMD will do okay, especially if you OC it. Safe bet would be to first upgrade to the AC cooler or for better OC go with the Zalman. You should be able to at least match the intel processor speed with an overclock on the AMD. If your budget can be stretched to the Intel processor, it would be a better choice if you treat each similarly (stock vs stock or oc vs oc). Either way, if you wanna overclock, upgrade your cpu cooling.
